The Causal Association of Sarcopenia-related Traits with Cardiovascular Disease: A Two-sample Mendelian Randomization Study

Objective: To explore the causal association between sarcopenia-related traits and common types of cardiovascular disease (CVD) by Mendelian Randomization (MR) analysis. Methods: MR analysis was performed using the Genome-Wide Association Study (GWAS) of the UK Biobank (UKB) for sarcopenia-related traits: appendicular lean mass (ALM), low hand-grip strength (LHGS), and usual walking pace (UWP), and the common types of cardiovascular diseases. Meanwhile, we used three different types of MR analysis methods: including Inverse Variance Weighting (IVW) method, MR-Egger Regression (MR-Egger) method, and Weighted Median method.
Results: Our study found that sarcopenia-related traits were associated with coronary atherosclerosis (ORALM = 0.909, 95% CI: 0.851-0.971, P-value = 4.46E-03; ORUWP = 0.387, 95% CI: 0.238-0.631, P-value = 1.39E-04), heart failure ( ORALM = 1.099, 95%CI: 1.048-1.152, P-value = 8.76E-05; ORLHGS = 0.860, 95%CI: 0.772-0.958, P-value = 0.006; ORUWP = 0.319,95%CI: 0.220-0.462, P-value = 1.63E-09), atrial fibrillation and flutter (ORUWP = 0.344, 95%CI: 0.193-0.614, P-value = 3.10E-04), any stroke (ORALM = 0.927, 95%CI: 0.887-0.970, P-value = 9.20E-04; ORUWP = 0.515, 95%CI: 0.344-0.769, P-value = 0.001) and valvular heart disease (ORALM = 1.101, 95%CI: 1.052-1.152, P-value = 2.91E-05) were partially causally associated in the association analysis. However, our findings do not provide significant evidence of a causal association between peripheral arterial disease and ALM, LHGS, and UWP. Conclusion: This study provides further valid evidence on the causal association between sarcopenia-related traits and common types of CVD. In the future, it is still necessary to pay attention to the problem of sarcopenia-related traits in CVD patients, which may be helpful in providing early warning signs and effective interventions for them.
